If you have recently upgraded to WordPress 3.0 and facing problems while upgrading, do check out the sticky that has been created in WordPress forums regarding WordPress 3.0 Issues, Problems, Resolutions. So far, problems have been reported regarding TSG podcasting plugin, Headspace2 plugin, events-calendar plugin error, allowed memory size exhausted error and unavailable for scheduled maintenance error. Some people also reported blank page after upgrade while some are not able to edit the pages from dashboard.
So if you facing any of such similar issues, my suggestion is DON’T PANIC! Take a look at these threads. There are great many people ready to help you out. Though most of the issues are related to incompatible plugins, some might be related to the way WordPress was upgraded. Many people reported issue with automatic upgrade (though I didn’t face any such issue luckily). If nothing works, contact your host and ask them to switch back to the older version of WordPress.
